ABSTRACT

Objective To analyze the clinical effects of internal radiation of iodine-125 seed implantation in the treatment of advanced pancreatic carcinoma.Methods 22 patients with unresectable and advanced pancreatic carcinoma were treated by the internal and interstitial radiation using iodine-125 seed implantation.Results Survival of 20 months was observed in 5 patients,10 ~ 20 months in 9 patients,2 ~10 months in 8,patients with an average of (13.47 ± 8.12) months.Among the 22 patients,complete response was obtained in 5 patients,partial response in 8 patients,no response in 7 paticnts,and the other showing PD.The response rate( CR + PR) was 59%.Conclusions Intra - operative internal radiation by iodine - 125 seed implantation does show some therapeutic effects for advanced pancreatic carcinoma.

ABSTRACT

Objective To study the significance of CD24 protein expression in esophageal squamous cell carcinoma and its relationship with clinical pathological parameters and clinical prognosis.Methods The expression of CD24 protein was studied in 90 esophageal squamous cell carcinoma samples by EnVision immunohistochemical analysis and to study relationship of CD24 with clinical pathological parameters and prognosis.Results The expression of CD24 in esophageal squamous cell carcinoma was significantly higher than that in adjacent normal tissue (x 2 =0.832,P < 0.05).The expression of CD24 was not related with the age of patien,sexuality and the histologic grade (x2 =0.741,0.361,0.930,8.858,all P > 0.05),but with clinical stage of the cance (x2 =10.804,P =0.005),lymph node metastasis (x2 =12.349,P =0.000) and depth of tumor invasion (x2 =13.94,P =0.007).Significant difference in survival between the different CD24 expression group,the OR (95 % CI) values were 2.105 (61.022-69.273),2.313 (55.108-64.176),1.286 (32.388-37.429),the survival of patients with strong expression of CD24 was inferior to that of weak expression of CD24,depth of tumor invasion and lymph node metastasis were independent prognostic factors of esophageal squamous cell carcinoma.Conclusion CD24 may be related to the malignant process of esophageal squamous cell carcinoma.Overexpression of CD24 indicates poor prognosis in esophageal squamous cell carcinoma patients.CD24 may become a new independent prognostic factor for esophageal squamous cell carcinoma.

ABSTRACT

Objective The goal of the study was to assess the lethal effect of cytotoxic lymphocyte against A549 cells induced by dendritic cells (DC) pulsed with A549 lysate and transfected with GM-CSF recombinant adenovirus. Methods The cytotoxic lymphocyte against A549 cells were induced by culturing with DCs, which pulsed with A549 antigens and transfected with GM-CSF recombinant adenovirus. The of effector/target ratio, the killing rates of N-DC group, A-DC group and G-A-DC group were (1.9±0.7) %,effector/target ratio, the killing rates of N-DC group, A-DC group and G-A-DC group were (5.3±0.2) %, (40.5±7.7) % and (72.5±4.7) %, respectively. We found that the killing rate of G-A-DC group was the highest by statistics. Conclusion The cytotoxic lymphocyte against A549 cells can be induced by DCs pulsed with A549 lysate ,and the lethal effect of CTLs can be enhanced when DCs were infected with GM-CSF recombinant adenovirus.

ABSTRACT

OBJECTIVE To study 40 kinds of resistant-related genes in a pan-resistant Pseudomonas aeruginosa.METHODS To detect the susceptibility of antimicrobial agents by MIC,40 resistant-related genes including 29 ?-lactamases genes,porin oprD2 genes,6 aminoglycoside-modifying enzymes(AMEs)genes,chlorhexidine/sulfadiazine resistant gene(qacE△1-sul1)and intergron(intⅠ1,2,3),etc,form 1 strain of P.aeruginosa were measured by PCR,and verified by DNA sequencing.RESULTS In the strain,there were positive of 6 kinds of resistant-related genes(blaTEM,blaOXA10,aac(6′)-Ⅱ,aac(3)-Ⅱ,qacE△1-sul1 and intⅠ1),but without oprD2 genes.Twenty-seven kinds of ?-lactamases genes,4 kinds of AMEs(aac(6′)-Ⅰb,aac(3)-Ⅰ,ant(3″)-Ⅰ and ant(2″)-Ⅰ),and 2 kinds of intⅠ(intⅠ2 and intⅠ3) were negative.CONCLUSIONS The multi-resistant mechanisms of pan-resistant P.aeruginosa are mainly related to 7 kinds of resistant-related genes(blaTEM,blaOXA10,oprD2,aac(6′)-Ⅱ,aac(3)-Ⅱ,qacE△1-sul1 and intⅠ1).

ABSTRACT

OBJECTIVE To study 6 kinds of aminoglycoside-modifying enzymes (AMEs) genes in Chryseobacterium spp isolates. METHODS The isolates were identified by API20NE Gram-negative identification cards,and the susceptibility of antimicrobial agents was detected by MIC kits ( bioM?rieux ) 6 AMEs genes of 2 strains of Chryseobacterium spp were measured by PCR,and verified by DNA sequencing and sequence analysis. RESULTS In the 2 strains,2 kinds of resistant genes [aac(6′)-Ⅱ and ant(2″)-Ⅰ] were positive,and 4 kinds genes of AMEs [aac (6′)-Ⅰb,aac(3)-Ⅱ,ant(3″)-Ⅰ and aac(3)-Ⅰ] were negative.The amplicons were purified,sequenced and analyzed with BLAST 2.0 and found to be identical to aac(6′)-Ⅱ and ant(2″)-Ⅰ. CONCLUSIONS There are AMEs in Chryseobacterium spp isolates. This is the first report on AMEs genes [coexistance of aac(6′)-Ⅱ and ant(2″)-Ⅰ] in Chryseobacterium spp.

ABSTRACT

Objective To investigate the expression of CD24 and FAK in the invasive breast ductal carcinoma tissues and their clin-ical significance. Methods Tissues were obtained from 86 patients with breast carcinoma and 30 non-cancer specimens. The expression of CD24 and FAK was detected by immunohistochemical technique (ElivisionTMPlus) and their correlation with tumor clinicopathological char-acters was analyzed. Results The positive rate of CD24 and FAK expression in 86 cases of breast carcinoma was 87.2% (75/86) and 72.1% (62/86) respectively, which was significantly different from that in control group(P<0.01). Expression of CD24 was significantly correlated with axillary lymph node metastasis(P<0.01). Expression of FAK was correlated with histopathologic grade, axillary lymph node metastasis and clinical stage (P<0.05). There was a positive correlation between the expressions of CD24 and FAK in breast carci-noma tissues (rs = 0.222, P < 0.05). Conclusion Expression of CD24 and FAK are closely correlated with the development of breast carcinoma and there is a markedly positive correlation between CD24 and FAK expression. These indexes can be helpful in understanding the biological behaviors and predicting the prognosis of breast carcinoma.

ABSTRACT

Objective To investigate the expression of CD24 and Ki67 in the invasive breast ductal car-cinoma tissues, so as to assess the role of CD24 protein in the carcinogenesis and progression of breast canc-er. Methods The expression of CD24 and Ki67, Bcl-2 was examined by immunohistochemically in 86 breast cancer specimens and 30 non-cancer specimens ( ElivisionTM Plus). The relationship between CD24 expression and Ki67, Bcl-2, patient age, tumor size, histopathologic grade, axillary lymphnode metastasis, and clinical stage of breast cancer was analyzed. Results In 86 eases of invasive breast ductal carcinoma, the positive incidences of CD24 was 87.2% (75/86), which distinguished apparently from those of the con-trol group( P <0.01 ). In breast carcinoma tissues, the expression of CD24 was positively related to axillary lymphnode metastasis(P <0.01 ), but was not related to patients age,tumor size, histopathologic grade and clinical stage(P>0.05). CD24 expression was positively related to Ki67 (P<0.01 ) and negatively relat-ed to Bcl-2(P>0.05). Conclusion CD24 expression was related to the proliferation, invasion and me-tastasis of breast cancer cells and it can serve as an important marker for predicting biological behavior and prognosis of tumors.

ABSTRACT

OBJECTIVE To investigate the antimicrobial-resistant genes in one strain of Acinetobacter baumannii isolated from the 1st Hospital,Quanzhou,Fujian Province,China.METHODS To detect the susceptibility of antimicrobial agents by MIC,29 kinds of ?-lactamases genes were analyzed by PCR and verified by DNA sequencing and sequence analysis.RESULTS The strain of A.baumannii according to the susceptibility to antimicrobial agents by MIC of the API PSE 5.0 antibiotic susceptibility cards(bioM?rieux) and BD Phoenix NMIC/ID-109 identification/antibiotic susceptibility cards was pan-resistant,2 kinds of genes of blaTEM and blaADC were positive,sequence analysis revealed that the 2 PCR products were the ?-lactamases,the blaADC was a new type(blaADC-Quanzhou,GenBank EU 200768).CONCLUSIONS There are the blaTEM and blaADC genes in the strain of A.baumannii isolated from the 1st Hospital,Quanzhou,Fujian Province,China,the blaADC is a new type.

ABSTRACT

Objective To study the expression of human platelet factor 4 (hPF4)mRNA and its relationship with angiogenesis and lymph node metastasis in papillary throid carcinoma(PTC). Methods RT-PCR was used to detect the expression of hPF4 mRNA in 40 cases of PTC and 10 cases of normal tissue surrounding carcinoma. Immunohistochemical stain was performed to detect microvessel density(MVD) whose endothelia were marked by antibody CD105. Results The positive-expression rate of hPF4 mRNA was 80 % respectively in normal tissue surrounding carcinoma. The positive-expression rate of hPF4 was 47.5 % in PTC cases. ABSTRACT

Objective To investigate the significance of the expression of interleukin-10(IL-10),interleukin-13(IL-13)and interleukin-15(IL-15)in serums of patients with hepatitis B.Methods The expression of IL-10,IL-13 and IL-15 in serums of 109 patients was measured by ELISA.Results The serum levels of IL-10,IL-13 in patients with moderate degree chronic hepatitis B(CHB)group were significantly higher than that in patients with chronic severe hepatitis group,severe degree CHB group,acute hepatitis group and normal group.The serum level of IL-15 was increased in patients with acute hepatitis、moderate degree CHB group,severe degree CHB group and chronic severe hepatitis compared with normotensives.The proportion of IL-15/IL-10 and IL-15/IL-13 in patients with chronic severe hepatitis group,severe degree CHB group and acute hepatitis group were higher than that in patients with moderate degree CHB group and normotensives.The serum level of IL-15 and the proportion of IL-15/IL-10 and IL-15/IL-13 in dead group were significantly higher than that in improving group with chronic severe hepatitis.Conclusion There is an abnormal cell-mediated immune response in patients with hepatitis B.Combining detection on the levels of serum IL-10,IL-13 and IL-15,and the proportion of IL-15/IL-10 and IL-15/IL-13 would nicely show the cellular active status and be helpful to the prediction of prognosis and direction of therapy in patients with hepatitis B of different kinds of clinical type.

ABSTRACT

OBJECTIVE To study resistant mechanisms in a pan-resistant Pseudomonas putida strain.METHODS To detect the susceptibility of antimicrobial agents by MIC,6 aminoglycoside-modifying enzymes(AMEs) genes of 1 strain of P.putida were measured by PCR,and verified by DNA sequencing and similarity searches of DNA sequencing data banks using BLAST.RESULTS In the strain,there were 4 positive kinds of AMEs resistant genes(aac(6′)-Ⅰb,aac(3)-Ⅱ,ant(3″)-Ⅰand ant(2″)-Ⅰ).Sequence analysis revealed that the 4 PCR products were AMEs,the aac(6′)-Ⅰb was a new type(GenBank EU 137667).CONCLUSIONS The mechanisms of resistance to aminoglycosides of the pan-resistant P.putida are mainly related to 4 kinds of AMEs,the aac(6′)-Ⅰb is a new type.

ABSTRACT

Objective: To study the probiotic properties of 12 selected strains of lactobacilli.Methods: The probiotic properties of 12 selected strains of lactobacilli were investigated for their resistance to pH 2.5 and 0.3% oxgall,their adhesion to Caco-2 cells and antimicrobial activity against enteric pathogenic bacteria.Results: The 12 strains all showed relatively high resistance to acid and bile salt and varied antimicrobial activities against pathogenic bacteria of intestinal infection.Lactobacillus johnsonii JCM1022 exhibited a broad inhibitory effect on enteric pathogenic bacteria,especially on enteropathgenic E.coli and Salmonella enteritidis,while Lactobacillus reuteri JCM1081 possessed a high activity of adhesion to Caco-2 cells(495.07?80.03).Conclusion: Among the 12 strains,Lactobacillus johnsonii JCM1022 and reuteri JCM1081 are qualified candidates for clinical application.

ABSTRACT

<p><b>OBJECTIVE</b>To make a thorough study on the clinicopathologic significance of the three-dimensional structural alteration of the p53 protein in papillary thyroid carcinomas and to provide an objective criterion for the evaluation of PTC prognosis.</p><p><b>METHODS</b>A total of 41 PTC cases were enrolled. Techniques including polymerase chain reaction with single strand conformational polymorphism (PCR-SSCP), DNA sequencing, computerized three-dimensional protein modeling by means of international shared resources and related software analysis were used.</p><p><b>RESULTS</b>15 cases with p53 gene mutation defined as Group I were detected in totally 41 PTC cases. No p53 gene mutation was found in the rest 26 cases which were classified as Group II. The differences in lymph node metastatic rate, distant metastatic rate, age, sex, size of the lesion between Group I and Group II were not significant (P > 0.05). The alterations of the amino acid residues of 9 PTC cases out of the 15 p53-gene mutated patients (Group I) were either located in the p53-protein domains, mainly the core domain and the non-specific DNA binding basic domain, or the severely defect cases with the formation of widely divergent structures. It was found that the alterations of the structure of the core domain could directly check the binding of p53 protein to its target DNA molecules. In addition, the alterations of the structure of the basic domain could indirectly prohibit the binding. The ones mentioned above were classified as Group Ib. The rest of six cases with their p53 protein amino acid residues mutated beyond the domains were grouped as Group Ia. The differences in lymph node metastatic rate, distant metastatic rate between Group Ib and Group Ia were statistically significant (P < 0.01, P < 0.05) respectively.</p><p><b>CONCLUSIONS</b>The alterations of the three-dimensional structure of p53-protein is considered as one of the morphological basis of the progression and heterogeneity of PTC. They render an authentic evidence for the selection of the clinical cases with a poor risk for metastasis.</p>

ABSTRACT

To determine the effect of pre-operative chemotherapy on apoptosis in breast cancer and to evaluate its signif icance as a prcgnostic marker. MethodsPatients with breast cancer were divided into preoperative chemotherapy group (40 cases)and control group (42 cases). Two groups were analyzed for the appearance of apoptosis by using TUNEL method and electron mi croscope in tissue sections. ResultsApoptosis occurred in 92.5 % of preoperative chemotherapy group and in 78.5 % of control group. The apoptotic indexes were 19.37 + 6.49 and 9.26 + 5.04 ( P < 0.01 ) respectively. Low apoptotic index was related to disease-free survival of patients with breast cancer (P < 0.01 ). ConclusionThe preoperative chemotherapy can induce apoptcsis of breast cancer and improve disease-free survival.

ABSTRACT

Objective To explore the value of pneumatic lithotripsy under ureteroscope in the management of ureteral calculus . Methods The clinical data of 360 cases of ureteral stones treated ureteroscopically by pneumatic lithotripsy were retrospectively analyzed. Results Stones were successfully fragmented on one session in 85% of the cases (306/360). Complications were observed in 2 5% of the cases (9/360), including 6 cases of ureteral perforation and 3 cases of urinary infection. Conclusions Pneumatic lithotripsy under ureteroscope is safe, effective and minimally invasive.
